How do I defend my asking price during negotiation?

Defend your anchor with data, not stubbornness. Present recent comparable sales that directly support your number and force the buyer to justify their lower bid against hard evidence. Watch for the midpoint rule - if you counter too low, you implicitly accept a new, lower midpoint. Keep your counter close to your asking price and let the comps do the negotiating for you.
